1. Excise band of interest from a TAE gel; estimate volume by weighing the gel slice.
2. Dissolve the gel slice in 3 volumes of NaI (from Geneclean kit) at 55EC for 5 min or until the gel dissolves.
3. Add 1 ml of Wizard miniprep resin, 10 min. at room temperature.
4. Add DNA resin mixture to a syringe and pass through a minicolumn.
5. Wash with 2 ml of Wizard wash buffer; spin 2 min in a microfuge.
6. Elute with 70EC TE or water (50 μl), 2 min in a microfuge.
