1. take 4 microliters of the peptide solution in water (1 g/l),
2. add 2 microliters of carboxypeptidase Y in water ( 0.01 g/ l), then
3. add 14 microliters of 5 mM NaAC buffer pH 4.0 at room temperature.

Comments

A sequencing ladder forms in 2 hrs. The ladder will be incomplete, but some information will usually be present .
