PREPARATION OF ANTI-BLEACHING MOUNTING MEDIUM
Materials
1. p-phenylenediamine, 1%(w/v) in distilled water, 1 ml. Adjust pH to 9.5 with NaOH. Use the highest grade from Sigma.
2. glycerol.
3. 10x simple PBS.
Procedure
1. Mix 1 ml p-pnenylenediamine, 1 ml 10x PBS, 6.9 g glycerol, and 3 ml distilled water.
2. Adjust pH to 7.8.
3. Divide into aliquots and store at -20oC. Protect from light.
4. The medium will gradually turn dark upon storage. It has to be replaced when the color turns dark purple.
5. Mounted slides can be stored at 4oC for several weeks if protected from light.
